 medications, the US Food and Drug Administration (FDA) granted several Orphan Drug and Fast Track designations to various drugs throughout June, from a hereditary hemorrhagic telangiectasia treatment to an oral drug for chronic neutropenia. Additionally, multiple drugs were successful in their respective studies, with iptacopan achieving its primary endpoint in a phase 3 trial and denecimig proving its tolerability in hemophilia A after switching from emicizumab.

VAS-101 (Vasceptor) Receives Orphan Drug Designation for Sickle Cell Disease

On June 12, 2025, the FDA granted Orphan Drug designation to VAS-101 for sickle cell disease. A patented topical curcumin formulation delivered through parent company Vascarta’s patented transdermal technology, VAS-101 is intended to fill the niche caused by limited bioavailability and effectiveness of oral curcumin dosing.

FDA Grants Fast Track Designation to Mavorixafor for Chronic Neutropenia

Mavorixafor, previously approved for the treatment of WHIM syndrome in 2024, has now received  Fast Track designation to treat chronic neutropenia, granted by the FDA on June 10, 2025. The only other medication approved for CN, a human recombinant granulocyte-colony stimulating factor, causes several side effects, such as thrombocytopenia, osteoporosis, vasculitis, and increased risk of leukemia. Mavorixafor aims to fill this void, as few patients receive the optimal dose recommendation of the G-CSF medication.

FDA Grants Orphan Drug Designation to DIAG723 for Treatment of HHT

Announced on June 18, 2025, the first-in-class antibody DIAG723, which aims to restore ALK1 signaling and promote vascular quiescence in patients with hereditary hemorrhagic telangiectasia, also received Orphan Drug designation from the FDA. Parent company Diagonal Therapeutics has begun a natural history study of adults with HHT to characterize variability of patient-reported outcomes such as epistaxis, hematologic support, and quality of life.

Genetically Modified Cell Therapy BEAM-101 Successful in Sickle Cell Disease Trial

Announced by parent company Beam Therapeutics at the European Hematology Association 2025 Congress, BEAM-101 has established a strong clinical profile. The one-time treatment, administered via a hematopoietic stem cell transplant, is designed to trigger increased production of non- and anti-sickling fetal hemoglobin.

Denecimig Well-Tolerated in Hemophilia A After Switching from Emicizumab

The phase 3b FRONTIER5 trial – fifth and final trial in the ongoing phase 3 FRONTIER program – demonstrated switching to denecimig from emicizumab can be done safely without a washout period or loading dose. Parent company Novo Nordisk also announced its intention to submit denecimig for US and EU regulatory review later in 2025.

Iptacopan (Fabhalta) Hits Primary Endpoint in Phase 3 APPULSE-PNH Trial

APPULSE-PNH aimed to investigate the safety and efficacy of Novartis’s twice-daily oral monotherapy iptacopan in adults with paroxysmal nocturnal hemoglobinuria. On June 12, 2025, Novartis announced that patients treated with iptacopan achieved Hb ≥12 g/dL and clinically meaningful fatigue improvements without requiring transfusion.

Articles

The FDA has granted several Orphan Drug and Fast Track designations in June 2025, despite no major hematology drug approvals. VAS-101 received Orphan Drug designation for sickle cell disease, while Mavorixafor was granted Fast Track status for chronic neutropenia. DIAG723 also received Orphan Drug designation for hereditary hemorrhagic telangiectasia. In research, BEAM-101 showed promise in sickle cell disease trials, denecimig was well-tolerated in hemophilia A, and iptacopan met its primary endpoint in a phase 3 trial for paroxysmal nocturnal hemoglobinuria.

Bimekizumab Demonstrates Long-Term Benefit for Arthritis Post-Approval, with Philip Mease, MD

Bimekizumab has demonstrated sustained improvements in disease control and symptoms for up to 3 years in people with psoriatic arthritis (PsA) and active axial spondyloarthritis.

These findings were presented in a several posters at the 

European Alliance of Associations for Rheumatology (EULAR) Congress 2025 

in Barcelona, Spain, which was held from June 11-14, 2025.

spoke with Philip Mease, MD, director of rheumatology research at Providence Swedish Medical Center and clinical professor at the University of Washington School of Medicine, Seattle, who served as an investigator on the studies of bimekizumab, to learn more about the new data presented and how it showcases the therapy’s benefits for both people with PsA and AxSpA.

Mease went over some highlights from the BE OPTIMAL and BE COMPLETE studies in people with PsA and the MOBILE-1 and MOBILE-2 studies in people with AxSpA. He emphasized the sustained efficacy seen across the 3 years in both populations, especially in minimal disease activity (MDA), which he considered to be of particular value as an end point. Furthermore, he went over the favorable safety profile seen with bimekizumab, which has some risk of adverse events (AEs), but which yielded low rates of AEs across the follow-up.

“MDA achievement was 53% at 3 years. This is really the highest number that I've seen for this important composite measure that's a target of treatment and so I thought that was a not only meaningful in terms of the height of response, but also the fact that it was still being maintained out at 3 years,” Mease said.

Bimekizumab was approved by the FDA for treating adults with active PsA, active non-radiographic AxSpA, and active ankylosing spondylitis in adults in September 2024.

On January 28, 2025, Novo Nordisk announced FDA approval of semaglutide (Ozempic) to reduce the risk of kidney disease progression, kidney failure, and cardiovascular death in adults with type 2 diabetes and chronic kidney disease (CKD). Based on the phase 3b FLOW trial, semaglutide reduced the risk of major kidney events by 24% compared to placebo. According to Novo Nordisk, this makes Ozempic the most broadly indicated GLP-1 RA in its class. The trial was stopped early for efficacy, showing additional benefits for cardiovascular outcomes. This approval expands treatment options for millions facing type 2 diabetes with CKD.

FDA Approves Suzetrigine, a Non-Opioid Option, for Treatment of Acute Pain

On January 30, 2025, Vertex Pharmaceuticals announced FDA approval of suzetrigine (Journavx), a first-in-class, non-opioid NaV1.8 pain signal inhibitor for moderate-to-severe acute pain. Based on results from Phase 3 trials in post-surgical patients, suzetrigine significantly reduced pain over 48 hours compared to placebo and showed comparable efficacy to opioids without associated risks like sedation or respiratory depression. According to Vertex, suzetrigine marks the first new pain treatment class approved in over 20 years and offers an effective alternative for opioid-sparing strategies. Notable Attributes of Icotrokinra for Adolescents with Psoriasis, with Lawrence Eichenfield, MD

In this interview from Q2, Lawrence Eichenfield, MD, discussed data released by Johnson & Johnson from a subgroup analysis of the ICONIC-LEAD study that had been presented at the 2025 World Congress of Pediatric Dermatology (WCPD) Annual Meeting. Eichenfield described the data that demonstrated that adolescents with moderate-to-severe psoriasis given once-daily icotrokinra (JNJ-2113) attained higher rates of clear or almost clear skin at the 16-week mark compared to placebo.

Roflumilast Foam 0.3% for Scalp, Body Psoriasis Effective, Safe for Patients

Another notable study from Q2 showed positive phase 3 results on the safety and efficacy of roflumilast foam 0.3% (Zoryve) as a monotherapy for patients with psoriasis of the scalp and body. These results, demonstrating rapid and significant relief following treatment, were the conclusion of the phase 3 ARRECTOR study, authored by such investigators as Melinda Gooderham, MD, MSc, medical director at the SKiN Centre for Dermatology. Gooderham and the coauthors expressed, following the analysis, that before their work, topical therapeutic options for scalp psoriasis had been limited by efficacy, by formulation, and/or by safety.

Baricitinib Confirmed Safe and Efficacious for Treating Alopecia Areata

Long-term findings from the integrated safety analysis of patients with severe alopecia areata in the BRAVE-AA1 and BRAVE-AA2 clinical trial program, spanning up to 4 years, were released and reinforced the established efficacy and safety profile of baricitinib over an extended period. Baricitinib, an oral selective Janus kinase (JAK) inhibitor, has been approved by the regulatory bodies of many different countries for several rheumatologic and dermatologic disorders, as well as for COVID-19.

Azathioprine Effective for Patients with Recalcitrant, Severe Alopecia Areata

 in Q2 demonstrated that azathioprine is an efficacious systemic treatment for individuals living with recalcitrant and severe alopecia areata, according to research authored by Susan Farshi and Parvin Mansouri, from the Skin and Stem Cell Research Center at Tehran University of Medical Sciences in Iran. Farshi and Mansouri’s study participants came from the dermatology clinic at Imam Hospital and the Skin and Stem Cell Research Center at Tehran University of Medical Sciences. The team's prospective study involved the tracking of patients living with alopecia areata over a 10-year timeframe.

Lebrikizumab Leads to Atopic Dermatitis Improvements in Patients with Skin of Color

New phase 3b findings on lebrikizumab (Ebglyss) released in Q2 highlighted the drug’s efficacy for atopic dermatitis in adult and adolescent patients with skin of color, as well as results in itch and disease severity improvements over 24 weeks. This late-breaking data resulted from the open-label ADmirable study and was presented at the 2025 Revolutionizing Atopic Dermatitis (RAD) Annual Meeting in Nashville. It was led by such authors as Andrew F. Alexis, MD, MPH, professor of clinical dermatology and vice-chair for diversity and inclusion at Weill Cornell Medicine. Alexis and colleagues had noted the lack of robust data prior to the ADmirable study informing the diagnosis and treatment of atopic dermatitis in patients with skin of color.

Upadacitinib Effective for Disease Clearance, Itch in Older Adults with Atopic Dermatitis

In new data presented at RAD 2025, older adults with atopic dermatitis aged ≥ 65 years treated with oral upadacitinib 15 mg as a once-daily monotherapy were shown to attain optimal skin clearance and relief from itch, along with notable quality of life and sleep improvements. These data on older adults ≥65 years with moderate-to-severe atopic dermatitis were the result of 140 weeks of evaluation in the phase 3 Measure Up 1 and 2 analyses.

DermaSensor Aids PCPs in Spotting Skin Cancer

Recent findings from Q2 in the pivotal DERM-SUCCESS study, along with a companion utility study, provided strong clinical support for DermaSensor, a handheld skin cancer assessment device that was cleared for primary care by the FDA in 2024. The DERM-SUCCESS analysis had assessed the diagnostic performance of DermaSensor, a noninvasive, elastic scattering spectroscopy device, for melanoma and keratinocyte carcinoma detection within primary care settings. This evaluation was carried out in 22 clinics within the US and Australia, with adult patients with skin lesions suspected of malignancy by primary care physicians (PCPs) being involved between August 2020 - December 2021.

Investigators Develop AI Model That Can Predict Nail Psoriasis

Investigators from the Friedrich-Alexander-University Erlangen-Nurnberg in Germany have developed a convolutional neural network, designed to automatically and objectively score the severity of patients’ nail psoriasis with high reliability and no need for image standardization. Psoriasis is known to often impact nails, given their categorization as dermal appendages. Additionally, there is an estimated total of 80-90% of individuals who eventually present with nail involvement who already have psoriasis.
